1.0 PURPOSE
The
purpose of this document is to describe the procedure
used when necropsying jirds for the collection of
Brugia sp. and to describe the procedure used when
transplanting adult Brugia sp. intraperitoneally
into recipient jirds.
2.0 PROCEDURE
2.1 Collection of Brugia sp.
2.1.1 Prior to necropsy, euthanize the jird(s) by
exposure to CO2.
2.1.2 Shave the abdominal area of the jird(s).
2.1.3
Examine the peritoneal cavity and remove the
Brugia sp. worms.
2.1.4 Dissect the heart, lungs, and testes and
remove the worms.
2.1.5 Place all of the worms from the jird(s) in TC
Hank=s
balanced salt solution containing an appropriate
antibiotic such as penicillin/streptomycin.
2.2 Surgical Transplantation of Brugia sp.Into the
Peritoneal Cavity
2.2.1 Administer pentobarbital sodium solution (1
grain/mL ) to the jird by intraperitoneal (IP) injection
at a rate of 0.5-0.7 mg/g b.wt. to produce general
anesthesia.
2.2.2 Prepare the jird(s) by clipping the right
ventral abdomen and swabbing the skin at that location
with 70% ethanol.
2.2.3 Make a small laparotomy incision and insert
the Brugia sp. IP using a sterile bulb pipette.
2.2.4 Close the laparotomy incision with an auto clip
staple (or suitable alternative).
2.2.5 Monitor the jird(s) during recover from
anesthesia.
