Characteristics of the
Brugia malayi microfilaria cDNA Library |
Please quote reference number SAW94LS-BmMf in the source section of
your Genbank submission form when you submit sequence data from this
library.
Vector: Lambda Uni-ZAP XR (Lambda ZAP cut with Eco RI and Xho I -
Stratagene)
Insert: Brugia malayi microfilaria cDNA.
Cloning Sites: EcoRI (5' end) and XhoI (3' end)
Unamplified Titer: 7.0 X 105 pfu/ml
Amplified Undiluted Titer: ~ 1010 pfu/ml
Average Insert Size: ~ 1100 bp
Background: < 10% non-recombinants
Host Strain: XL-1 Blue MRF' strain
Host Strain Information:
The XL-1 Blue MRF' strain is the recommended host strain for amplification
and screening of this Lambda Uni-ZAP cDNA library. The XL-1 Blue MRF'
strain is restriction minus and is recommended for plating unamplified
libraries, and for amplifying and screening libraries. It also enables
blue/clear color selection of recombinant clones when grown on plates
containing IPTG and X-gal.
The bacterial stock is stored and streaked on LB-tet plates and grown as
overnight cultures in the absence of tetracycline.
Strain Genotype:
Δ (mcrA) 183, Δ (mcrCB-hsdSMR-mrr) 173, end A1, supE44, thi-1, recA1,
gyrA96, relA1, lac [F' proAB, lac IqZΔ M15, Tn10 (tetr)].

Upon Receiving the Library:
The library is frozen in 7% DMSO. Thaw in hand quickly and dilute 1:10 in
SM buffer immediately. Add chloroform to 0.3%, mix, and store at 4oC.
